MAPK Signaling
Rat sarcoma (RAS) is an upstream protein in the mitogen-activated protein kinase (MAPK) signaling pathway1
RAS is activated by several stimuli that primarily involve the formation of complexes comprising autophosphorylated growth factor receptors, the adapter protein growth factor receptor–bound protein 2 (GRB2), and the exchange factor son of sevenless homolog 1 (SOS1)1
Specifically, the signaling adapter protein GRB2 facilitates the binding of receptor tyrosine kinases (RTKs) or anchoring proteins to SOS1/2, which activates RAS paralogs2
The activated SOS1 subsequently induces the guanosine diphosphate (GDP) and guanosine triphosphate (GTP) exchange of RAS paralogs and thereby activates the MAPK cascade2
SOS1 inhibition is a promising therapeutic strategy for diseases involving the dysregulation of the MAPK pathway3
