Compound Overview
Kirsten rat sarcoma viral oncogene homolog (KRAS)/mitogen-activated protein kinase (MAPK) pathway dysregulation is among the most frequent drivers of cancer.1 Son of sevenless homolog 1 (SOS1) is a guanine nucleotide exchange factor that facilitates the activation of KRAS by promoting the exchange of guanosine diphosphate (GDP) for guanosine triphosphate (GTP).2 Inhibiting SOS1 with SOS1 inhibitor (BAY 3498264) prevents KRAS activation, thereby disrupting downstream signaling through both the MAPK and phosphatidylinositol 3-kinase (PI3K) pathways.2-4
SOS1 inhibitor (BAY 3498264) is currently being evaluated in a phase I clinical trial involving patients with advanced KRAS G12C–mutated solid tumors.3
